Day 1: ELISA Fundamentals & Plate Preparation

    • Direct, Indirect, Sandwich, and Competitive ELISA

  • Role of antibodies, antigens, and enzymes

  • Common sources of error in ELISA experiments

Hands-On / Demonstration

  • Microplate coating

    • Antigen or antibody coating (real or mock setup)

  • Preparation of wash buffers and blocking solutions

  • Understanding plate layout and controls

Day 2: Antigen–Antibody Reaction & Detection

  • Antigen–antibody binding specificity

  • Signal generation and amplification

  • Importance of washing and incubation steps

Hands-On / Mock Experiments

  • Antigen–antibody reaction using mock or safe samples

  • Addition of detection antibody / enzyme conjugate

  • Color development step (demonstration-based)

  • Troubleshooting scenarios

    • High background

    • Weak signal

    • Inconsistent readings

Day 3: OD Measurement, Data Analysis & Interpretation

  • Preparation of standard curve

  • Calculation of unknown sample concentration
