3-Day Hands-On Workshop on Microbiology Techniques

This workshop introduces participants to fundamental microbiology laboratory practices, focusing on media preparation, aseptic handling, culturing techniques, staining, and microbial analysis. For safety and educational purposes, the workshop uses non-pathogenic, commonly encountered microorganisms.


Day 1: Media Preparation & Sterilization

  • Introduction to microbiology and microbial growth requirements

  • Types of culture media: simple, enriched, and selective (conceptual overview)

  • Principles of sterilization and contamination control

Hands-On Training

  • Preparation of culture media (e.g., nutrient agar)

  • Sterilization methods

    • Autoclave (if available)

    • Pressure cooker as an educational alternative

  • Pour plate technique

  • Safe handling of glassware and media

Day 2: Isolation Techniques Using Safe Cultures

  • Culturing microorganisms from safe sources

    • Curd / yogurt

    • Soil microbes (educational sampling)

  • Incubation and observation of microbial growth

Day 3: Staining, Enumeration & Microbial Analysis

  • Observation of colony characteristics

    • Size, shape, margin, elevation, and color

  • Colony Forming Unit (CFU) calculation

  • Recording and tabulating observations
